摘要
目的 :探讨长骨器官体外培养的最佳条件。 方法 :游离受孕第 16天小鼠胚胎尺骨在体外进行培养 ,观察培养基 p H值、血清含量和通气方式等因素对尺骨生长发育的影响。结果 :将尺骨置于 p H为 7.2~ 7.4、血清含量为 10 %~ 2 0 %的 BGJ( b)培养基 ,以连续通气或每 2 4小时通气一次的方式进行体外培养 ,可保证培养尺骨正常生长。 结论 :本研究为探索有害物质 (如氟等 )
Objective: To investigate the optimum of organ culture of long bone in vitro. Method: By means of the method of autorotation with constant temperature, ulnas from 16 day old mouse embryos were cultured in vitro. Results: The growth and development of bone were better under condition of pH 7.2~7.4, 10%~20% serum in the culture medium and continuous (or once a day) supplementation of gas mixture. Conclusion: This study provided a useful experimental condition for investigating the skeletal damage of toxicants.
