实验性急性心肌梗死心肌细胞凋亡与p53和Bcl-2基因mRNA表达的研究 被引量:2

Regulation of p53 and Bcl-2 gene on myocyte apoptosis in acute myocardial infarction in rabbit

摘要 目的 探讨p5 3和Bcl- 2基因在实验性急性心肌梗死 (AMI)心肌细胞凋亡中的作用。方法 用TUNEL法检测 48只兔AMI模型梗死区的心肌细胞凋亡 ,用以cDNA为内参标的逆转录 聚合酶链反应 (RT -PCR)的方法检测其心肌细胞内p5 3和Bcl- 2基因mRNA表达量 ,并探讨它们之间的相互关系。结果 TUNEL法发现兔梗死区心肌细胞凋亡千分率在冠脉结扎前( 0h)和结扎后 1h、 2h、 3h、 4h、 6h、 8h、 12h的改变如下 :0h组和 12h组 <1h组 <2h组和 8h组<3h组 <4h组 (P均 <0 0 5 ) ;p5 3基因mRNA表达量依次为 :冠脉结扎后 4h组 >结扎后 6h组>结扎后 3h组 >结扎后 2h组 >未结扎 ( 0h)组和结扎后 1h组、 8h组、 12h组 (P均 <0 0 5 ) ;且梗死区心肌细胞凋亡数与其内p5 3mRNA表达量对数值之间成明显的正相关性 (r为 0 930 0 ,P<0 0 0 1)。Bcl- 2基因mRNA表达量 :冠脉结扎后 3h组、 4h组和 6h组 <结扎后 2h组、 8h组 <结扎后 1h组和未结扎 ( 0h)组 <结扎后 12h组 (P均 <0 0 5 ) ;且梗死区心肌细胞凋亡数分别与其内Bcl- 2mRNA表达量的对数值成明显的负相关性 (r为 - 0 8732 ,P <0 0 0 1) ;p5 3和Bcl- 2基因mRNA表达量的对数值之间亦呈明显的负相关性 (r=- 0 96 49,P <0 0 0 1)。结论 急性心肌梗死时心肌细胞存在明显的凋? Objective To investigate the expression and significance of p53 and Bcl-2 gene in myocyte apoptosis in infarct area in the AMI rabbit model.Methods TUNEL was applied to monitor the myocyte apoptosis in 48 rabbits.p53 and Bcl-2 gene mRNA expressions were assessed by RT-PCR using cDNA sample as an internal standard,and their relationship was also investigated.Results In the AMI rabbit model,TUNEL method showed that the permillage of apoptotic myocytes in infarct area at oh(before) and 1h,2h,3h,4h,6h,8h,12h after coronary artery occlusion were:0h group and 12h group 1h group<2h group and 8h group<3h group<4h group(P<0 05).The amounts of mRNA expression of p53 gene were:4h group>6h group>3h group>2h group>0h group>1h group,8h group,12h group after coronary artery occlusion(All P<0 05) A significantly positive correlation was found between the number of apoptotic myocytes and logarithmic value of p53 mRNA expression(r=0 9300,P<0 001).The degrees of expression of Bcl-2 mRNA were:3h group,4h group and 6h group<2h group,8h group<1h group and 0h group<12h group after coronary artery occlusion(All P<0 05).A significantly negative correlation between the number of apoptotic myocytes and logarithmic value of Bcl-2 mRNA expression(r=-0 8732,P<0 001) were found.Furthermore,the logarithmic value of p53 mRNA and Bcl-2 mRNA expression was signifcantly negatively correlated(r=-0 9649,P<0 001).Conclusions Distinct characteristics of apoptosis in myocytes of infarct area in the AMI rabbit model were found,which were up regulated by p53 gene and doun-regulated and Bcl-2 gene.
