摘要 以牛血清白蛋白(BAS)和牛血清红蛋白(HG)为物系,在制备型高效液相色谱(HPLC)分离装置上,考察了操作方式对柱效和分离度的影响。结果表明,在制备分离过程中,在满足分离要求的前提下,可以采用柱超载方式操作,以提高制备量。但当一次操作所要处理的样品量较小时,采用质量超载方式较为有利。当一次操作所要处理的样品量太大,导致分离效果不能满足分离要求或样品在流动相中的流动度达到极限时,采用体积超载方式更为有利。 The effect of operating method on column efficiency and resolution was investigated by using the preparative HPLC with the mixture of bovine serum albumin and bovine blood hemoglobin. The results show that if the separation demand can be satisfied,column can be overloaded in order to achieve more preparative throughput. However,if the amount of sample in one operation is very small, mass overload method would be better. If the amount of sample in one operation is so large that the separation demand can t be satisfied or the solubility of the sample in the mobile phase reaches to the upper limit, volume overload would be better.
