降压合剂对胰岛素抵抗高血压大鼠作用效应及机制的实验研究 被引量:16

摘要 目的 :观察降压合剂对胰岛素抵抗高血压大鼠的作用效应 ,探讨其作用机制。方法 :雄性 SD大鼠饲以高糖高盐饲料制模 ,然后分组给予不同药物灌胃 ,观察各组大鼠体重、收缩压 (SBP)、空腹血糖 (FBG)、空腹血胰岛素 (FINS)、胰岛素敏感性指数 (ISI)、红细胞 Na+ K+ ATP酶及血管紧张素 (Ag )的变化。结果 :(1)高糖、高盐饲料喂大鼠 4周时 SBP、FINS升高 ,ISI降低 (P均 <0 .0 1) ,FBG无变化。 (2 ) 8周时 ,与正常组比较 ,模型组红细胞 Na+ K+ ATP酶活性降低 ,Ag 升高 (P均 <0 .0 5 )。与模型组比较 ,用药各组 SBP、FBG、FINS下降 ,ISI升高 (P <0 .0 5或 P <0 .0 1) ;高降压合剂组、卡托普利组、降压合剂 +卡托普利组红细胞Na+ K+ ATP酶活性升高 (P均 <0 .0 5 ) ,卡托普利组 Ag 降低 (P<0 .0 5 )。结论 :高糖高盐饲料喂养大鼠能产生高血压及胰岛素抵抗。降压合剂能降压、降糖、降血胰岛素、增加胰岛素敏感性。降压合剂的降压及增加胰岛素敏感性的作用可能是通过增高 Na+ K+ ATP酶活性这一途径。 Objective:To observe the effects of Jiangya mixture(JYM,降压合剂) on insulin resistance hypertensive rat model and investigate its mechanism.Methods:Male Spraque Darley rats were fed on high glucose and high salt diet to establish experimental model,following by giving different drugs to divided groups.The changes in body weight(BW),systolic blood pressure(SBP),fasting blood glucose(FBG),fasting blood insulin(FINS),insulin sensitivity index(ISI),red blood cell(RBC) Na+K+ATPase,angiotensinⅡ(AgⅡ) were observed.Results:(1)At fourth week,SBP and FINS were increased but ISI decreased in rats fed on high glucose and high salt diet(all P<0.01).(2)At eighth week in comparison with normal group the activity of RBC Na+K+ATPase was decreased but AgⅡ increased (all P<0.05)in model group.In Comparison with model group the SBP,FBG and FIN[HJ5”:2]S were decreased,otherwise ISI was increased (P<0.05 or P<0.01).In other groups,meanwhile the activity of RBC Na+K+ATPJYM+captopril group and AgⅡ decreased (P<0.05) in captopril group.Conclusions:Hypertension and insulin resistance can be induced in rats by feeding high glucose and high salt diet.The JYM can reduce blood pressure,blood sugar,and level of blood insulin otherwise increase insulin sensitivity.The mechanisms of it on reducing blood pressure and increasing insulin sensitivity may be obtained by heightening the activities of Na+K+ATPase.
