摘要
目的为寻找适合临床应用的新型MDR逆转剂,探讨了鸦胆子油乳剂对人胃腺癌长春新碱耐药细胞MKN28/VCR的影响作用。方法采用MTT法进行体外耐药逆转实验,免疫细胞化学染色法显示耐药细胞中MRP蛋白的分布。结果分别合用丁基硫堇亚胺和鸦胆子油乳前后,MKN28/VCR对抗癌药物阿霉素、顺铂、丝裂霉素、5-氟尿嘧啶、依托泊甙和长春新碱的敏感性均有显著性差异(各组都为P<0.01);经鸦胆子处理后的耐药细胞中MRP蛋白的表达明显减少。结论鸦胆子油乳剂对人胃腺癌长春新碱耐药细胞的耐药性有逆转作用。
AIM To screen new reversals of drug resistance, the effect of emulsion of seminal oil of brucea javanica on vincristine-resistant human gastric cancer cell lines was studied. METHODS The reversing drug resistance was studied by MTT and the expression of protein associated with multidrug resistance were examined by immunocytochemistry. RESULTS MTT cytotoxic assay revealed that the sensitivities of MKN28/VCR cells to ADM, CDOP, MMC, 5-Fu, Vp16 and VCR were different according to presence or absence of buthionina sulphoximina and emulsion of seminal oil of brucea javanice (P<0.01, respectively). The immunocytochemistry showed that after treated with emulsion of seminal oil of brucea javanica, MRP staining was reduced in MKN8/VCR cells. CONCLUSION The drug resistance of MKN28/VCR cells could be reversed by emulsion of seminal oil of brucea javanice.
