摘要
目的 探讨Galardin对角膜基质细胞胶原降解的抑制作用。方法角膜基质细胞三维胶原凝胶表面分别添加纤维蛋白溶酶原、白细胞介素-1α(IL-1α)及不同质量浓度的 Galardin的 MEM培养液培养 24 h,测定培养上清液中羟脯氨酸的量做为胶原降解活性单位。向时检测 Galardin及 IL-1α对角膜基质细胞胶原降解的作用。结果在存在纤维蛋白溶酶原的情况下,随着 IL-1α质量浓度的升高,以剂量关系增加角膜基质细胞胶原降解量。 Galardin以剂量关系抑制 IL-1α诱导的角膜基质细胞胶原降解量。结论Galardin具有抑制 IL-1α诱导的角膜基质细胞胶原降解的作用。
ObjectiveTo study the inhibitive effect of Galardin on keratocyte collagen degradation.Methods Keratocyte collagen gels were incubated with plasminogen,interleukin-1α(IL-1α)and different concentration Galardin MEM culture medium for 24 hours separately.The ultrafiltrates were measured then hydrolyzed,and the amount of hydroxyproline were measured spectrophotometrically.The effect of a synthetic matrix metallaproteinase(MMP)inhibitor,Galardin,and interleukin-1α (IL-1α)on collagen degradation were also examined.ResultsIn the presence of plasminogen,IL-1α increased keratocyte collagen degradation in a dosedependent fashion.Galardin suppressed IL-1α-induced collagen degradation more strikingly in a dose dependent fashion.ConclusionGalardin inhibites keratocyte collagen degradation induced by IL-1α.
