摘要
目的 探讨诱导气道上皮细胞NO产生的分子机制。方法 采用培养的人支气管上皮细胞系研究了TNF α及IL 1β对气道上皮细胞NO终末产物硝酸盐和亚硝酸盐产生的影响。结果 培养的支气管上皮细胞到达单层时加入TNF α和IL 1β ,其硝酸盐及亚硝酸盐水平均较对照组明显增高 (P <0 .0 0 1)。TNF α浓度 5 μg/ml和IL 1β浓度10 μg/ml时 ,刺激气道上皮细胞NO产生达高峰。时间曲线研究发现TNF α1和L 1β刺激气道上皮细胞产生NO的作用随时间增加而增强 ,在 3小时达高峰。结论 研究表明气道上皮细胞是气道内源性NO产生的主要细胞之一 ,TNF α和IL 1β在气道上皮细胞NO产生中起重要作用。
Objective To explore the molecular mechanisms of nitric oxide production in airway epithelial cells.Methods Human bronchial epithelial cells line(BEAS 2B)treated with ⅠL 1β or TNF α were studied by comparison with control.Nitrite and nitrate were assayed with colorimetric assay Kit(Cayman Chemical).Results Nitrite and nitrate were significantly higher in cultured human bronchial epithelial cells with IL 1β and TNF α than those in control( P <0.001).Cultured human bronchial epithelial cells with IL 1β or TNF α resulted in a time dependent and dosage dependent increase in nitrite and nitrate,compared with bronchial epithelial cells cultured in media alone.Conclusion NO is induced in bronchial epithelial cells following the treatment of cytokines IL 1β and TNF α.The results of the present study demonstrate that IL 1β and TNF a play an important role in NO production of human airway epithelial cells.
