摘要
目的研究碱性成纤维细胞生长因子(bFGF)对局灶脑缺血的作用。方法采用线栓法制备右侧大脑中动脉闭塞局灶性脑缺血模型。术后即刻分别腹腔注射bFGF和生理盐水。测定缺血前后血压、心率,缺血前、缺血后30分钟~24小时的局部脑血流量(rCBF),缺血24小时血清神经元特异性烯醇化酶(NSE),观察缺血24小时病理学改变和行神经功能评分。结果发现bFGF可降低血清NSE,改善组织病理和rCBF,促进神经功能恢复,但不影响血压、心率。结论bFGF能减轻缺血性脑损害,保护脑组织。
Objective To study the effect of basic fibroblast growth factor (bFGF) treating focal cerebral ischemia.Methods The animal models of focal ischemia of the right middle cerebral artery(MCA) occlusion was established by suture method.bFGF (100μg/kg) in vehicle or vehicle(n=5, respectively) alone was injected intraperitoneally after MCA occlusion.We investigated blood pressure and heart rate before and after ischemia, serum NSE at 24 hours after MCA occlusion,neurological deficit score,regional cerebral blood flow(rCBF) 30 mimutes to 24 hours before and after ischemia and pathological changes.Resaults bFGF reduced serum NSE,improved pathology and rCBF,and promoted recovery of neurological function,but no effect on blood pressure and heart rate. Conclusion bFGF can reduce ischemic brain damage and protect cerebral tissue.
