摘要
甜樱桃 (Prunusavium)和酸樱桃 (Prunuscerasus)分步微繁技术 ,此技术涵盖了微繁全过程所需的全套技术 ,包括外植体和培养基的制备方法 ,各个阶段的培养技术及培养基配方、试管苗炼苗和移栽等 ,采用这一技术体系 ,可在 30周内完成从芽外植体建立到温室植株的全过程。此外 ,还提出了进一步改进该技术体系的建议 ,如调整总氮量及NO3-/NH4 +比率 ,添加蔗糖以外的其他碳源 。
This paper describes a step-by-step protocol of micropropagation for sweet cheery (Prunus avium) and sour cherry (P. cerasus) . The protocol covers the full information related to the whole process of micropropagation, including preparation of initial explants and media, a detailed account of the culture techniques and medium formulae, and hardening and transplanting of the micro-plants. By using the protocol, complete plants of greenhouse acclimatization can be regenerated within 30 weeks from bud explants. To further improve the protocol, some suggestions are offered, including adjusting the total nitrogen content and the ratio of NO 3- to NH 4 +, supplementing sugars other than sucrose and optimizing the concentrations of BAP and IBA and their balance.
