摘要
目的 :采用凝胶排阻色谱法测定尿多酸肽注射液中的小分子肽的分子量。方法 :采用Waters 5 10型高效液相色谱仪 ,TSK -GELG2 0 0 0SW×L色谱柱 (30 0mm× 7 8mm ,粒度 5 μm) ,以乙腈 -水 -三氟乙酸 (10∶90∶0 0 5 )为流动相 ,流速 :1 0mL·min- 1 ,紫外检测器于 2 14nm测定。结果 :测定尿多酸肽注射液中的小分子肽的分子量均为 10 4Da以下。结论 :方法简单 ,重现性好 ,可作为测定小分子肽分子量的参考依据。
Objective:To establish a HPLC method for the determination of peptide's molecular weight in Niaoduosuantai injection.Methods:A Waters 510 model HPLC instrument with TSK-GEL G2000 SW×L(300 mm×7.8 mm,5 μm) was used.The mobile phase was a mixture of acetonitrile-water-TFA(10∶90∶0.05),at a flow rate of 1\^0 mL·min -1and the ultraviolet wavelength was set at 214 nm.Results:The molecular weight of peptide in Niaoduosuantai injection was no more than 104 Da.Conclusion:The method is simple,reliable and accepted by the specification for controlling the quality of Niaoduosuantai injection.
