摘要
目的 探讨前列腺癌组织中端粒酶hTRT基因的表达及意义。 方法 应用原位分子杂交技术 ,对 47例前列腺癌 (PCa)、2 1例良性前列腺增生 (BPH)和 10例正常前列腺 (NP)组织中端粒酶hTRT基因进行检测和定位 ,并运用图像分析系统对hTRT表达水平与PCa分化程度的相关性进行研究。 结果 端粒酶hTRT基因在PCa中表达阳性率为 93.6 % ,表达强度与肿瘤细胞的分化程度显著相关 :未分化腺癌 >低分化腺癌 >中分化腺癌 >高分化腺癌 ;癌旁组织中hTRT基因表达率为 8.5 % ,在肿瘤组织和癌旁组织中差别有极显著性意义 (P <0 .0 1)。端粒酶hTRT基因表达水平与肿瘤细胞的分布定位一致 ,NP组织及BPH组织中端粒酶hTRT基因表达均为阴性。 结论 原位杂交检测端粒酶hTRT基因对PCa细胞水平的定位诊断具有重要意义 。
Objective To study the expression of hTRT gene in prostatic adenocarcinima. Methods Using in situ hybridization(ISH) techniques, the expression of telomerase gene hTRT mRNA in 47 prostatic adenocarcinoma、 21 BPH tissues and 10 nomal prostatic tissues were observed. Results The expression of hTRT was significantly higher in prostatic adenocarcinoma than in paracancer tissue( P < 0.01 ). There was a positive correlation between the expression of hTRT and the location of tumor cells. The density of telomerase hTRT expression was also correlated with tumor cell differentiation:undifferentiated >low defferentiated >moderately defferentiated >highly defferentiated).In nomal prostatic tissues and BPH tissues, telomerase hTRT was not detected. Conclusions The detection of hTRT was valuable to clinical oncology diagnosis and may provide a new approach for the gene therapy of prostatic adenocarcinoma.
