氟化钠对小鼠卵母细胞成熟和体外受精的影响 被引量:2

摘要 目的 探讨氟化钠对小鼠生殖功能的影响。方法 采用小鼠卵母细胞体外培养、体外受精的方法研究了氟化钠对小鼠卵母细胞成熟和受精能力的影响。结果  2 0 0mg/kg组氟化钠可以显著降低小鼠超排卵数 ,10 0mg/kg组和 2 0 0mg/kg组均可以显著抑制卵母细胞第一极体的释放 ,降低体外受精率 ,但对小鼠生发泡破裂没有影响 ,对卵母细胞的体外存活率也无显著影响 ;本实验结果还显示 ,2个染毒组均可以抑制体外受精 2 4h时的受精卵的卵裂 ,但是随着在正常培养液中培养时间的延长 ,这种抑制作用逐渐解除。结论 氟化钠可以破坏卵母细胞的成熟 ,降低卵母细胞的受精能力 。 Objective To study the reproductive toxicity of sodium fluoride in experiment systems. Methods The effect of sodium fluoride on meiotic maturation and fertilization capacity of mouse oocyte was studied by in vitro culture of mouse (treated by sodium fluoride) oocyte and in vitro fertilization (IVF). Results The results showed that 20 0 mg/kg bw of sodium fluoride by oral administration reduced the number of oocytes of superovulation 10 0 mg/kg bw and 20 0 mg/kg bw of sodium fluoride inhibited the extruding of the first polar body of mouse oocyte and reduced the rate of IVF, but little impact on germinal vesicle breakdown (GVBD) and viability of mouse oocytes The result also showed that 2 sodium fluoride treated groups exhibited inhibition of zygotes cleavage in IVF at 24 h At the same time the restrain effect may be gradually vanished by the culture of mouse zygotes in normal culture solution (CZB). Conclusion The results indicated that sodium fluoride may be hazardous to the meiotic maturation of mouse oocyte and reduces fertility Therefore, sodium fluoride has obviously reproductive toxicity
