摘要
目的 研究晚期混合裂变产物内照射诱发Wistar大鼠外周血淋巴细胞次黄嘌呤鸟嘌呤磷酸核糖转移酶(HPRT)基因突变情况。方法 将晚期混合裂变产物自尾静脉注入 ,再用 5 -溴脱氧尿嘧啶 (BrdU)法测定HPRT基因的突变频率。结果 随着内照射剂量的增加 ,大鼠外周血淋巴细胞HPRT基因突变频率也不断上升 ,内照射剂量在 10 .178cSv以上的各组与对照组均有显著差异 (P <0 .0 1)。剂量效应关系符合线性模型。结论 BrdU法是一种快速、简便、较准确的测定放射性核素内照射损伤的方法 ,HPRT基因突变可作为一种很好的生物剂量计。
Objective To study the relation between HPRT gene mutation of rat peripheral blood lymphocytes and the dose of internal exposure to late fission products.Methods The late fission pro ducts were injected into the bodies of some rats through the tail vein,then some time later,using the method of BrdU labeling which has been established,we got the mutant frequencies of the HPRT gene of the rats.Results With the increment of the accumulative dose,the mutant frequencies of the HPRT gene rose accordingly.Except the first exposed group,the others were significantly different from the control respectively. There is a linear relation between the radiation doses and the HPRT gene mutant frequencies.Conclusion According to the results of the research,it can be concluded that the BrdU labeling is a good method to study the mutation of the HPRT gene of somatic cells and the HPRT gene mutation can be used to calculate the dose of radiation.
