摘要
目的 观察电针足三里穴对胃粘膜细胞受体表达及核增殖的影响 ,探讨针灸作用机制 .方法 75只 SD大鼠随机分段分为 5组 ,在清醒状态下针刺实验 ,SABC法免疫组化染色观察胃粘膜细胞胃泌素 (Gas)、表皮生长因子受体(EGFR)表达程度 ,Ag NOR特染观察胃腺细胞增殖情况 .结果 电针足三里穴胃粘膜 EGFR阳性表达率 73.3% ,Gas表达率 6 6 .7% ;电针三阴交对 EGFR表达无影响 ,Gas表达率为 46 .7% ;联合穴位组 Gas,EGFR表达均达 6 0 .0 % ,Ag-NOR特染各组核内嗜银颗粒无明显差异 .结论 电针足三里可提高胃粘膜细胞 Gas和 EGFR表达率 ,改变细胞分子结构 ,影响细胞功能 .
AIM To observe the effect of electroacupuncture stimulation of zusanli on the expression of Gas and EGFR in gastric mucosa in order to probe mechanisms of acupuncture. METHODS 75 SD rats were randomized into 5 groups: Control group, non point group, zusanli group, sanyinjiao group and unite point group. All rats were experimented on consciousness. Immunohistochemical staining by means of SABC or SP technique was used to observe the expression, location and distribution of Gas and EGFR in gastric mucosa. Meanwhile, gastric mucosa cells were specially stained with AgNOR. RESUITS EGFR and Gas in gastric mucous tissue of the zusanli group after EA showed a positive expression significantly higher than those of the control and the non acupoint group. The expressions of Gas and EGFR were enhanced significantly (66.7% and 73.3%, respectively) in zusanli group. The expression of Gas in gastric mucous tissue were enhanced significantly (46.7%), but the expression of EGFR kept stable in Sanyinjiao group. The expression of Gas was enhanced as significantly as that of EGFR in zu san groups. Besides, nuclear argentaffin particles that were specially stained with AgNOR didn't change significantly. CONCLUSION Electroacupuncture stimulation of zusanli can enhance the expression rate of EGFR and Gas in gastric mucoas, but exert no effects on the proliferation of gastric mucous cells.
