甘蓝型油菜种皮色泽及相关性状的研究 被引量:21

摘要 本文研究了三套不同来源的黑籽和黄籽甘蓝型油菜近等基因系种皮中的总酚、花色素、类黄酮、黑色素含量及其相关的多酚氧化酶、苯丙氨酸解氨酸、酪氨酸酶活性在种子发育过程中的动态变化。实验结果表明 ,种皮中总酚、花色素及类黄酮在种子发育的前期逐渐增加 ,在开花 35至 4 3天后 ,其含量开始下降 ,PPO和 PAL活力与总酚和花色素的变化趋势一致 ;黑色素呈单调上升趋势 ,尤其是在种子成熟时 ,它在黑籽种皮中的含量迅速增加 ,是同时期黄籽油菜的 6倍多 ;酪氨酸酶也与黑色素的变化趋势一致。方差分析表明 ,多酚、类黄酮、花色素和黑色素在黑籽与黄籽种皮中的差异达到了极显著水平。在初期和中期 ,黑籽与黄籽色素含量的差异主要是与多酚、类黄酮和花色素有关。而在种子发育的后期 。 Three pairs of near isogenic lines for black and yellow seedcoat in B.napus were used as experimental materials and dynamic changes in the contents of polyphenols, flavonoid, anthocyanin and melanin and the activity of their related enzymes (polyphenol oxidase (PPO), phenylalanin ammonialyase (PAL) and tryosinase) in their seed coats were investigated in different developmental stages. Results showed that the contents of polyphenols, anthocyanin and flavonoid during the early stage of seed developing were increased gradually, and began to decline from 35~43 days after flowering. Melanin content increased sharply only near to seed mature, especially in black seed coat. Changing tendency of the activity of PPO and PAL was similar to that of polyphenols and anthocyanin, and that in tyrosinase activity was similar to that of melanin. Results also showed that during the early and mid stages the contents of flaonoid, polyphenols and anthocyanin in black seeds were much higher than those of yellow ones which was up to highly significant difference level, but there was no difference in melanin content. Conversely, during the late stage the content of melanin in black seed coat was more than 6 fold higher than that of yellow one for the corresponding period, but no difference in polyphenols, flavonoid and anthocyanin. It was speculated that color difference was mainly determined by polyphenols, anthocyanin and flavonoid in the early and mid stages, and finally by melanin in the late stage.
