摘要
通过农杆菌介导法用含NPT -Ⅱ和Barnase基因导入到大豆幼胚 ,大豆子叶节中 ,然后经过含Kan的筛选培养基中连续筛选 ,获得一批Barnase转基因植株。经PCR扩增结果 ,证实此基因已整合到大豆中 ,在当代植株中生物学特性观察。花粉粒不育率在 98%。
Npt-II and Barnase gene was introduced into soybean(Glycine max(L.)Merr.)immature cotyledonous and cotyledon nodes by gene transformation mediated by Agrobacterium tumefaciens. Kanamycin resistant(km R) plantlets were obtained by 3-4 cycles screening in selective condition .The presence of banase gene in the transgenic plants were confirmed by PCR.Results of PCR analysis proved that T-DNA was integrated into the genome of soybean. Alterant plants positive for barase gene,the pollen grains in complete male sterile were 98%.Factors influencing agrobacterium-mediated soybean transformation were investigated
