银染PCR-SSCP法检测52例白血病患者p53基因点突变 被引量:2

摘要 目的 通过检测白血病患者p5 3基因点突变 ,了解其与白血病的发展、治疗及预后的关系。方法 用银染PCR SSCP法检测 5 2例不同类型白血病p5 3基因第 5、6、7、8外显子的点突变。结果 在 45例急性白血病患者中有 5例发生p5 3基因突变 ,其中 3例对化疗不敏感 ;6例慢性粒细胞白血病中 1例有p5 3基因突变 ,此例患者在住院 1个月内死亡 ,1例毛细胞白血病发生p5 3基因点突变。结论 p5 Objective To investigate the relationship between the mutation of p53 gene and the carcinogenesis,development and prognosis of patients with leukmia.Methods We examined the mutation of p53 gene in exon 5,6,7,8 in 52 patients with leukemia by silver staining PCR SSCP.Results Mutation of p53 were found in 7 of 52 patients:mutation of p53 gene were found in 5 of 45 acute leukemia patients,among them three patients showed a poor response to chemical therapy;In 6 chronic myelocytic leukemia patients one was found the mutation of p53,she was died after one month since she had entered the hospital;one hairy cell leukemia patient was found the mutation of p53.Conclusion The mutation of p53 gene is prognostic factor for survival,it is instructive for clinical therapy.
