高碘、低碘对亲代及仔一代小鼠甲状腺形态定量体视学的影响 被引量:5

摘要 目的 研究高碘、低碘对亲代及仔代小鼠甲状腺形态定量体视学的影响。方法 用随机区组实验设计方法随机分为低、适、高碘三组,均饲低碘饲料,饮用含碘量分别为Oμg.L-1,50μg,L-1,3000μg.L-1,亲代鼠喂养100d,一部分动物处死取甲状腺,一部分生仔继续在相同条件下喂养100d,取甲状腺用HPIAS-1000图文分析系统测定甲状腺滤泡及滤泡腔体视学参数:平均面积(S)、面积体积(VA)、周长体积(VC)、比表面(S/V)、面数密度(NA)、形状因子(SF),并计算了甲状腺滤泡上皮细胞S、V占整个滤泡的百分比。结果1.低碘组亲代甲状腺滤泡S、VA、VC明显增大,滤泡腔S、VA、VC、SF、NA明显减小,S/V明显增大,而高碘组滤泡VC增大,滤泡腔S、VA、VC、SF增大,S/V、NA减小;2.低碘组仔一代鼠滤泡S/V、NA减小,滤泡腔S、VA、VC、NA明显减小,S/V增大;而高碘组滤泡S/V增大,滤泡腔的S、VA、VC增大,S/V、NA减小;3.低碘组亲代及仔一代滤泡上皮细胞面积、体积百分比明显增大,而高碘组则明显减小。结论 低碘组小鼠甲状腺滤泡上皮细胞明显增生肥大,滤泡腔明显变小。 Obiective To study the quantitative morphological stereology effect on thyroid in mice with high iodine and iodine deficiency. Methods The mice were divided into three groups by randomized block design,three groups were low iodine(LI),middle iodine(MI) and high iodine(HI) group. All mice were fed with iodine deficient forage, and were fed for 100 days with distilled water including different iodine doses, 0μg. L-1 (LI),50μg. L-1(MI), 3 000μg. L-1 (HI) respectively. The thuroid of 100d parental and 100d postnatal mice were fetched, and the thyroid follicle and folliculal cavities were measured with HPIAS(High Resolution Pathological Image & Word Analysis System). The stereology parameters included mean surface(S),volume on area(VA), volume on circumference(VC), specific surface(S/V), numerical density on area(NA), spherical factor(SF), the percentage of S and V of the follicular epithelial cell in thyroid follicle was further calculated. Results ① The thyroid follicle S,VA,VCin LI group were increased, the follicular cavities S,VA, VC, SF, NA were lower, S/V was increased, but the follicle VC in HI group was increased, the follicular cavities S, VA,VC, SF were decreased, S/V NA were increased. ② The follicle S/V, NA of postnatal mice in LI group decreased, the follicular cavities S, VA, VC, NA were decreased, S/V increased, but the follicle S/V of postnatal mice in HI group were increased, the follicular cavities S,VA,VC increased, S/V, NA decreased. ③ The follicular epithelial cell S, V percentage of parental and postnatal mice in LI group was increased, but that in HI group was decreased. Conclusion The follicular epithelial cell in LI mice were hyperplastic and plump, the follicular cavity significantly deflated, but the follicular cavity in HI geoup were significantly expanded, the follicular epithelial cell became thallus. The stumous degrees in postnatal mice reproduced by parental mice of high iodine and low iodine mice raised in the same condition was not more serious than that of parental mice.
