摘要
本研究采用RT PCR方法 ,以IFN γ和IL 2代表Th1型细胞因子 ,IL 4、IL 6、IL 10代表Th2型细胞因子 ,分别检测 3种肺癌细胞株 (鳞、腺、小细胞肺癌 )以及荷瘤机体外周血单个核细胞 (PBMC )Th1/Th2状态 ,并观察中药川芎嗪 (Tetram ethlpyrazine ,TTMP )的作用。结果显示 3种肺癌细胞株IFN γ和IL 2均无表达 ,而IL 4、IL 6、IL 10均表达 ;经川芎嗪培育后 ,3种肺癌细胞株均出现IFN γ的表达 ,而IL 4、IL 6的表达被抑制 ,小细胞肺癌株同时表达IL 2。肺癌患者PBMC中IL 4、IL 6和IL 10表达阳性率明显高于正常人 ,而IL 2无 1例表达 (0 / 2 3) ,IFN γ仅 1例表达 (1/ 2 3) ;与川芎嗪培养后 (两种浓度 ) ,肺癌患者IL 2 ,IFN γ的表达率明显提高 ;Th2型因子表达的优势状态较未加药组出现明显逆转。
To study the pattern of Th1/Th2 cytokines expression in human lung cancer cell lines and the peripheral blood mononuclear cells (PBMC) from lung cancer patients,and the effect of tetramethlpyrazine (TTMP) on them,three kinds of human lung cancer cell line and PBMC from 23 lung cancer patients as well as 19 healthy controls before and after the culture with TTMP were collected and the Th1/Th2 cytokine expression was determined The results showed that none of the human lung cancer cell lines expressed IL 2 and IFN γ,whereas IL 4,IL 6,IL 10 were expressed in pre cultured and control groups The expression of IFN γ in cultured groups was induced,while that of IL 4?IL 6 was inhibited,and the mRNA expressing rate of IL 4,IL 6,IL 10 in PBMC from lung cancer patients was obviously higher than that from the healthy controls IL 2 expression was not detected in all patients (0/23),and the positive rate of IFN γ expression was 1/23;After being cultured with TTMP,the expression of IL 4,IL 6,IL 10 were inhibited significantly In addition,the rate of IL 2 expression was enhanced significantly,indicating that TTMP could convert the imbalance state of Th1/Th2 cytokines expression in human lung cancer and make it incline to the normal state
