摘要
目的 建立一个合理、简单、并可验证的地塞米松屏蔽垂体的实验模型。方法 两组大鼠地塞米松 6mg·kg-1连续两次在两天内sc,对照组sc等容量的生理盐水。末次给药 1h后经地塞米松处理的一组ig受试药物Z47,另一组和生理盐水对照组ig等容量的 0 5 %CMC ,随即各组均ip0 0 86mol·L-1醋酸造成疼痛应激反应 ,以大鼠肾上腺VitC含量的降低作为衡量ACTH分泌的指标。结果 NS CMC HAc组VitC含量明显低于Dex CMC HAc组 ,差异非常显著 (P <0 0 1) ,表明地塞米松组垂体ACTH的分泌被完全阻断 ;Dex Z47 HAc组VitC含量低于Dex CMC HAc组 ,差异显著 (P <0 0 5 ) ,表明Z47对去垂体大鼠肾上腺仍有激动作用。结论 地塞米松处理的大鼠去垂体模型成立 ;
AIM To develop a simple reasonable and provable model of hypophysis shut off in rats with dexamethasone. METHODS Dexamethasone 6 mg·kg -1 was injected subcutaneously into two groups of rats at 3:00~3:30 pm on the preceding day. This dose was repeated at 9:00~9:30 am next day. The control group was treated with saline in the same way. One hour later, one of the dexamethasone treated groups was ig with Z47 200 mg·kg -1 , the other dexamethasone treated group and the control group was ig with 0 5% CMC. Then all the groups were ip with 0 086 mol·L -1 HAc in order to generate stress reaction. The reduction of the content of Vit C in adrenal gland was taken as the index of secretion of ACTH. RESULTS The content of Vit C in NS CMC HAc was obviously lower than in Dex CMC HAc group. There were statistically significant differences ( P <0 01), which indicted that the secretion of ACTH in dexamethasone treated groups was completely blocked. The content of Vit C in Dex Z47 HAc group was lower than in Dex CMC HAc. There were statistically differences ( P <0 05), which showed that Z47 had direct effect on the adrenals of the rats. CONCLUSION The model of hypophysis shut off by dexamethasone in rats is successful. Z47 can excite the adrenals of rats directly.
