摘要
目的 观察复方人参制剂对热应激鼠的保护作用。方法 3 4只SD大鼠被随机分为对照组 (Ⅰ组 ,n =17)、复方人参制剂组 (Ⅱ组 ,n =17)。热应激前 1h分别以水及复方人参制剂 (0 5ml/ 10 0g体重 )灌胃。动物于人工气候室内受热直至死亡。测定直肠温度、下丘脑hsp70含量等指标。结果 (1)体温的变化 :Ⅱ组动物热应激过程肛温各时点的测定值均较对照组低 ,其中热至 40min时低 0 18℃ ,热至 60min、 80min、 10 0min时则分别低 0 5 6℃、 0 70℃、0 71℃ (P <0 0 5 )。Ⅰ、Ⅱ组肛温的平均上升速率分别为 (0 0 44± 0 0 2 2 )℃ /min、 (0 0 2 9± 0 0 15 )℃ /min (P <0 0 5 )。(2 )动物热耐受时间和死亡率 :Ⅰ、Ⅱ组动物的平均热耐受时间分别为 (162 8± 62 9)min、 (2 17 4± 5 9 3 )min ,后者比前者延长 5 4 6min (P <0 0 5 ) ;其动物死亡率 ,在热暴露 2小时、 3小时、 4小时时Ⅰ组分别为 41 2 %、 5 2 9%和82 4% ,Ⅱ组则分别为 11 8%、 2 3 5 %和 5 2 9% ,P值均小于 0 0 5。 (3 )下丘脑hsp70含量的变化 :Ⅰ、Ⅱ组动物分别热暴露至死亡时 ,其下丘脑hsp70的含量为 (47 72± 19 0 3 ) μg/mg、 (5 4 3 2± 18 5 4) μg/mg (P <0 0 5 ) ,且hsp70含量与动物的热耐受时间呈高度正相关 (r
Objective To study the protective effects of compound ginseng preparation on the rats with heat stress.Methods Thirty four SD rats were allocated randomly into treatment and control groups,respectively,with 17 each.Compound ginseng preparation(0 5ml/100g body weight)and water were fed for the two groups,respectively,one hour before heat stress.And,the animals stayed at an artificial heat chamber until their death.Their rectal temperature and level of heat shock protein 70(hsp 70)in the hypothalamus were measured.Results The rectal temperature was 0 18℃,0 56℃,0 70℃ and 0 71℃ lower in the treatment group than that in the control group at 40min,60min,80min and 100min in heat stress,respectively.The average rate of temperature increase was (0 044±0 022)℃/min and (0 029±0 015)℃/min in the treatment group and control group,respectively.The length of heat tolerance was (162 8±62 9)min and (217 4±59 3)min in the treatment and control groups,respectively.Death rate of the animals was 41 2%,52 9% and 82 4% and 11 8%,23 5% and 52 9% in the treatment and control groups at two,three and four hours of heat stress,respectively,with a P value less than 0 05.The level of hsp 70 in the hypothalamus was(47 72±19 03)μg/mg and(54 32±18 54)μg/mg in the treatment and control groups,respectively,with a P value less than 0 05.Level of hsp 70 highly correlated with the length of heat tolerance,with a coefficient of correlation of 0 7074 and P<0 001.Conclusions Compound ginseng preparation could protect the animals from heat injury.
