摘要 目的建立超抗原诱导 T细胞失能的体外实验模型。方法 SEA刺激人外周血淋巴细胞 ,再加入外源性的 r IL-2维持 ,建立短期 SEA反应性 T细胞系 ;进行 Ficoll- Hypaque密度梯度离心获取经 SEA多次刺激过 SEA反应性 T细胞 ;加入处理过的 PBMC作 APCs,建立超抗原 SEA诱导 T细胞的活化组和失能组。结果 SEA和 r IL - 2共同作用可得到静息 SEA反应性 T细胞系 ;SEA反复刺激使 T细胞处于一低反应状态 ,与活化组相比 ,其 DNA合成能力明显下降 ,IL - 2分泌显著减少 ,静息一段时间后 ,其增殖能力慢慢恢复。结论超抗原 SEA多次刺激诱导 SEA反应性 T细胞处于失能状态。 ObjectiveTo establish a model of T cells anergy induced by superantigen in vitro. MethodsPBMCs were stimulated by SEA for 72 hours and then maintained with exogenous rIL-2 for two weeks to obtain resting transient SEA-reactive T cells line;Through Ficoll-Hypaque centrifugation, anergic T lymphoblasts stimulated repeatedly by SEA were obtained. ResultsIt was shown that resting transient SEA-reactive T cells line could be obtained through SEA and rIL-2 stimulation;T cells stimulated by SEA became unresponsive repeatedly. Contrast to SEA activated T cells, DNA synthesis and IL-2 production in anergic T cells appeared to be significantly reduced,but its proliferation could be restored after resting for a short time. ConclusionT lymphocytes stimulated by SEA repeatedly become anergic in vitro.
