摘要
目的 探讨改良型EC液低温灌注犬肺离体组织内一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量和湿干重比值(W/D)的变化,旨在为实验动物供肺保存研究提供分子水平的观察依据。方法 4 ℃改进型EC液灌注6只健康成年杂种犬肺并离体低温保存,30、60、120、180、240 min后取材测定不同时间NO、MDA、SOD含量和W/D,以灌注前为对照组。结果 不同时间点NO、SOD和MDA含量及W/D实验组与对照组之间无显著性差异(P>0.05);随时间延长NO、SOD均有下降趋势,MDA、W/D轻度上升。结论 改良EC液具有增加渗透压和清除氧自由基的作用;灌注前肺动脉内注射PGE1及增加代谢底物均有助于抑制供体肺保存期间氧自由基的产生。
: Objective To observe the changes of nitric oxide (NO), superoxide dismutase (SOD), malondialdehyde (MDA) and wet to dry weight ratio (W/D) in the canine pulmonary tissues perfused with improved Euro-Collins solution (iECS), providing a molecular insight into the preservation of the donor lung. Methods Six dogs were anesthetized to perform pulmonary perfusion with iECS at 4 ℃ and the isolated lungs were stored at 4 to 8 ℃ for 0, 30, 60, 120, 180, 240 min respectively, and the tissue samples of donor lung were subsequently taken for the measurement of NO, SOD, MDA contents and W/D at the appointed time points. Results The comparison of control and experimental groups did not reveal any significant differences in NO, SOD, MDA contents and W/D(P>0.05), but it was noted that NO and SOD contents tended to drop while MDA level and W/D rose as the duration of preservation was lengthened. Conclusion Mannitol can not only increase osmotic pressure but also help clearout oxygen-free radicals the production of which may also be inhibited by PGE1 injection prior to the perfusion and increased glucose content in EC.
