摘要
为了克服锥虫抗原变异对宿主免疫反应的干扰 ,探索伊氏锥虫病高效保护性抗原 ,根据伊氏锥虫在宿主体内第一次发生变异产生的变异抗原免疫原性相同的规律 ,设计了伊氏锥虫变异前抗原 (VSG1)和第二次变异所产生的抗原 (VSG2 )复合物作为免疫原 ,对小鼠进行免疫与单用变异前抗原免疫鼠相比较 ,两组鼠都用带变异前抗原的虫攻击 ,另以未免疫的鼠作对照 ,结果 VSG1+ VSG2免疫组小鼠 8只全部获得保护 ,单用 VSG1免疫组小鼠10只和未免疫组小鼠 10只血中全部出虫而死亡 ,前者比后者出虫时间和存活时间延长。提示运用伊氏锥虫抗原变异这一规律设计的这种免疫复合物 。
In order to overcome the disturbance on immunoprophylaxis by antigenic variation in Trypanosoma evansi and explore means of making the antigen which will be very useful to immunoprophylaxis of trypanosomosis, a complex antigen including VATs of ShTat1.1 and ShTat1.2 Trypamosoma evansi was produced according to regularity of antigenic variation in Trypanosoma. The compound immunoprophylaxis test with these antigen was carried out, mice were divided into three groups, one (10 mice) received antigen free, another (10 mice) received ShTat1.1 VSG alone, the third (8 mice) received both ShTat1.1 VSG and ShTat1.2 VSG, All mice were challenged with 100 ShTat1.1 Trypanosomes. The mice non vaceinted or vaceinated with ShTat1.1 VSG were all died on 6 th day and 9 th day respectively, Trypanosomes were discovered in tail blood of mice on 3th day and 6th day respectively, But all mice immunized with two VSGs were survived, It is suggested that immunoprophylaxis test designed according to some regularity of antigenic variation in T.evansi may give stronger protective effect.
