摘要
目的 研究煤焦沥青烟提取物致大鼠肺泡巨噬细胞 (AM )DNA损伤作用。方法 将大鼠肺泡巨噬细胞分 4组与不同浓度的煤焦沥青烟提取物共育 1h后 ,采用单细胞凝胶电泳技术检测大鼠肺泡巨噬细胞DNA断裂情况。结果 单细胞凝胶电泳后 ,对照组肺泡巨噬细胞无明显细胞拖尾现象 ,而经不同浓度的煤焦沥青烟提取物处理后 ,AM细胞有明显的拖尾现象 ,且随煤焦沥青烟提取物浓度增加 ,AM细胞彗星率上升 ,各组间差异有高度显著性 (P <0 .0 0 1) ,而且损伤的严重程度与作用物浓度呈相关关系 (Spearman相关系数为 0 .6 73,P <0 .0 0 1)。结论 煤焦沥青烟提取物作用于大鼠肺泡巨噬细胞可引起DNA链的断裂 。
Objective To study the DNA damage of alveolar macrophage induced by extracts of coal tar pitch fume in rat. Methods The alveolar macrophages (AM) were divided into 4 groups. These cells were exposured to 2.5?μg·ml -1 , 5.0?μg·ml -1 and 10.0?μg·ml -1 extracts of coal tar pitch fume and a control group. After incubating 1 hours, the single cell gel electrophoresis(SCGE) were used to detect the DNA strand breaks of AM. Results After undergoing electrophoresis, the 'comet' cells were seldom in control group, but the 'comet' cells increased significantly in experimental groups. There were significant differences in all groups(P<0.001). Conclusions Coal tar pitch fume extracts can induce DNA single strand breaks in alveolar macrophage of rat, and SCGE is suitable for the detection of DNA strand breaks in alveolar macrophage of rat.
