摘要
目的 建立贯叶金丝桃提取物及其原料中金丝桃素和伪金丝桃素的含量测定方法。方法 高效液相色谱法 ,其色谱条件是 :HypersilC18柱 ( 15 0mm× 4 .6mm ,5 μm) ;流动相为磷酸盐缓冲溶液 -甲醇 ( 3 5∶65 ,pH 6.0 ) ;流速为 0 .8ml/min ;检测波长为 5 90nm ,用外标法计算。结果 在 0 .0 6~ 0 .3 μg范围内金丝桃素和伪金丝桃素都呈线形关系 (r >0 .999) ,金丝桃素和伪金丝桃素的回收率分别是 98.6% (RSD =0 .81% ,n =5 )和 97.9% (RSD =1.0 2 % ,n =5 )。结论 建立的含量测定方法可用于贯叶金丝桃提取物整个生产过程中的质量控制。
WT5”BZ] Objective To establish a practical method for determining the hypericin and pseudohypericin in hypericum perforatum . Method By RP-HPLC,[column,Hypersil C 18 (150 mm×4.6 mm,5 μm);mobile phase,phosphate buffer solution methanol (35∶65,pH 6.0);flow rate,0.8 ml/min;wavelength,590 nm],quantity was calculated based on the external standards of hypericin and pseudohypericin. Results The linear relationship of calibration curves was good in the range of 0.06~0.3 μg for both hypericin and pseudohypericin,( r >0.999).The recoveries of hypericin and pseudohypericin were 98.6%(RSD=0.81%, n =5)and 97 9%(RSD=1 02%, n =5)respectively. Conclusion The method is simple,rapid, accurate and can be applied to quality control for the total producing process of ST.John's Wort extract.
