摘要
目的 探讨以生物活性聚己内酯 /聚乳酸共聚物 (PCLPLA)为载体的兔骺软骨细胞体外培养的可行性。方法 应用具有三维多孔立体结构的生物活性支架材料 (PCLPLA)为细胞外基质替代物 ,体外进行兔骺软骨细胞培养。采用组织学、电镜、MTT比色法、免疫组化法等多种手段动态检测细胞生长情况。结果 兔骺软骨细胞生长良好 ,增殖活跃 ,合成大量软骨基质 ,并与材料形成细胞———材料复合体 ,细胞充满于材料孔隙内。结论 生物活性可降解吸收材料PCLPLA具有良好的组织相容性及表面活性 ,它所提供的三维立体空间结构有利于细胞生长繁殖。为探讨骨骺早闭的全新疗法奠定了基础。
Objective To discuss the possibility of taking the new biodegradative and absorbable materials PCL/PLA as the carrier to cultivate epiphyseal plate cells in vitro in order to lay a foundation for the new method of treatment of growth arrest.Methods Carrying on the culture of epiphyseal plate cells in vitro by applying 3D and porous activable PCL/PLA as scaffolds,which are inside the body of organism and can be degraded and absorbed.Checking up the conditions of the growth of the cells by applying the ways of histology,electron microscope,MTT test and immunohistochemistry.Results The cells of epiphyseal plate grow well,increase actively,synthetize cartilage matrix and produce cells by combining the materials-cells compound,which the hole of the materials being full cells.Conclusion The newly biodegradative and absorbable materials PCL/PLA has the characters of better histocompatibility and surface activity.The 3D construction of that is propitious to the growth of the cells.The test lays a foundation for the new method of the treatment of growth arrest.
