摘要
目的 从细胞水平观察泰素帝对卵巢癌细胞株HO 8910的抑制作用及对分泌转化生长因子α、β的影响。 方法 细胞增殖抑制实验检测不同浓度的泰素帝对人卵巢癌细胞株HO 8910增殖的抑制率 ;放免法测泰素帝作用后TGF α的变化 ;ELISA法测泰素帝作用后TGF β1的变化。结果 泰素帝在一定浓度范围对HO 8910细胞增殖有明显抑制作用 ;泰素帝作用 48小时后 ,TGF α水平降低 ,TGF β1水平升高 ;TGF α与细胞增殖呈正相关 ,TGF β1与细胞增殖呈负相关。结论 泰素帝对人卵巢癌细胞有明显的抑制作用 ,在一定范围内呈量 效关系 ;正性生长因子TGF α及负性生长因子TGF β1可能参与泰素帝对卵巢癌细胞的作用机制。
Objective To investigate the regulatory effect of the secretion of transforming growth factor-α and β 1(TGF-α、TGF-β 1)by Docetaxel on ovarian cancer HO-8910 cell line and on cell proliferation in vitro.Methods The cytotoxicities were assayed with MTT;the secretion of TGF-α was investigated by using RIA kit;the secretion of TGF-β 1 was investigated using TGF-β 1 human quantitive ELISA kit.Results Docetaxel,at the concentration of 1×10 -4 ~1μg/ml,inhibited the growth of HO-8910 cell line obviously while IC50 was 6.31×10 -3 μg/ml;When treated with docetaxel for 48 hours,the secretion of TGF-α decreased and the secretion of TGF-β 1 increased.Conclusion The inhibition of docetaxel on HO-8910 cell line is great and gives dose-dependent responses;Docetaxel could inhibit the cell proliferation by down-regulating the TGF-α autocrine and up-regulate the TGF-β 1 autocrine.
