摘要 目的 :改进大鼠短暂性 ( <2 h)局灶性脑缺血模型的制作方法。方法 :采用雄性 SD大鼠 2 4只 ,实验组分别用线栓法致大脑中动脉 ( MCA)缺血 15、3 0、60 min,每组 6只 ;假手术对照组 6只。所有实验动物均采用乙醚麻醉 ,在手术过程中用 5 - 0丝线结扎颈外动脉 ,在结扎的近侧剪断颈外动脉或其分支——枕动脉 ,自颈外动脉或枕动脉断端将 4 - 0外科单丝尼龙线经颈总动脉分叉部插入颈内动脉直至大脑中动脉起始处。假手术对照组线栓插入颅内但不达大脑中动脉起始处。术后即观察动物行走时是否转圈。结果 :所有实验组动物都及时观察到转圈的表现 ,符合实验的入组标准。假手术对照组动物术后未观察到转圈的表现。结论 :短暂性局灶性脑缺血模型制作方法经改进更易于制作、可操作性更强 。 Objective:To modify the method of preparing transie nt(within 2 hours) focal cerebral ischemia model in rats.Methods:The middle cerebral artery(MCA)of male Sprague Dawley rats( n = 24)was occluded for 15,30 and 60 minutes( n =6 per group) using an intralumina l mon ofilament and six rats were served as shamoperated control group. All the rats w ere anesthetized with ether, and the external carotid artery was ligated with 5 0 silk suture during the operation. The proximal part of external carotid arte ry or occipital artery was cut with scissors and a 4 0 monofilament nylon sutur e was inserted into the lumen of external carotid artery or the occipital arter y th rough the cut end. The nylon suture was then gently advanced from the external c arotid artery to the internal carotid artery lumen till it passed into MCA. But the nylon suture did′t reach the MCA in the sham operated control group. Obser v ation was made immediately after operation to see whether the rat ran round in t he field or not.Results:All the rats ran round in the field immediately except the sha m operated control group.Conclusion:The modified method of preparing transient focal cerebral i schemia model in rats is easier to carry out than any ofter method used before.
