褪黑激素对吗啡戒断小鼠前列腺和睾丸治疗作用的酶组织化学观察

摘要 目的 观察褪黑激素 (melatonin ,MT)对吗啡成瘾小鼠前列腺及睾丸治疗作用的酶组织化学变化。方法 设立正常对照组 (A)和实验组 (B、C、D) ,实验组分别皮下注射定量吗啡 ,建立吗啡依赖模型 ,A组皮下注射等量生理盐水 ,而后B组继续注射吗啡 ,C组自然戒断 ,D组自然戒断后用MT灌胃。实验后将前列腺及睾丸作HE染色 ,光镜观察 ,并进行酶组织化学染色和图像分析。结果 吗啡依赖组小鼠前列腺和睾丸组织有不同程度萎缩 ,ACP活性增加 ,SDH活性降低 ;吗啡戒断组小鼠前列腺组织有明显增生 ,睾丸组织有轻度增生 ,ACP和SDH活性均增加 ;吗啡自然戒断后给MT组小鼠前列腺和睾丸组织结构基本正常 ,ACP和SDH活性均正常。结论 MT对吗啡依赖小鼠的前列腺和睾丸有治疗作用 ,可抑制吗啡戒断小鼠前列腺和睾丸组织的增生 。 Objective To observe the histological and enzymy histochemistry effect of melatonin on prostate and testis in mice of morphine dependent.Methods A physical dependent model in mice was established by subcutaneous injection of morphine. the histology and enzyme histochemistry changes on prostate and testis could be observed with microscope, and using enzyme histochemistry and imaging analysis the activity changes of major enzymes were observed.Results The prostate and testis in morphine dependent mice is atrophic and the activities of ACP were increasing while the SDH were decreasing; A multiplication of prostate could be observed in natural morphine withdrawal mice and the activities of ACP and SDH were increased; multiplication of prostate and testis could be inhibited by MT and the activities of ACP and SDH recovered normal.Conclusion The multiplication of prostate in morphine withdrawal mice could be inhibited by melatonin and the activities of ACP and SDH could be recovered normal.
