摘要
目的 :探讨 CD45/ SSC设门方法在儿童急性白血病流式细胞仪免疫表型分析中的优越性。方法 :采用CD45/ SSC设门的方法确定原始细胞群 ,与异硫氰酸荧光素和藻红标记的系相关抗体联合进行三色白血病免疫表型分析 ,并与 FSC/ SSC设门方法进行比较。结果 :CD45/ SSC设门与 FSC/ SSC设门比较能将骨髓原始细胞与正常淋巴细胞、单核细胞及粒细胞明显分开 ,所得原始细胞比例与形态学分类明显相关 ,免疫表型分析假阳性发生率低。 CD45与异氰酸荧光素和藻红蛋白标记的系相关抗体同时标记 ,能对白血病细胞群进行多参数分析 ,更好地反应白血病细胞抗原表达特性。结论 :采用 CD45/ SSC设门 ,能将骨髓中原始细胞与正常的淋巴细胞和单核细胞明显分开 ,假阳性发生率低 ,免疫分型结果可靠。
Objective:To investigate superiority of flow cytometry CD 45 /SSC gating for pediatric acute leukemia.Methods:Blast cells and antigen expression relating to leukemia were identified by flow cytometry CD 45 /SSC gating and the results were compared to FSC/SSC gating procedure.Rusults:This procedure uses the combination of CD 45 intensity and SSC to distinguish leukemic cells from normal lymphocytes,monocytes,neutrophils.The percent of blasts in correlated highly with morphologic leukemic cell counts compared with that of FSC/SSC,the ratio of false postive samples is low.When CD 45 was conbined with other pairs of fluorescein isothiocyanate (FITC) and phycoerythrin (PE) conjugated reagents,it is possible to analysis on leukemic blasts and display dual parameter dats regarding expression. Conclusion:The procedure of CD 45 /SSC gating can distinguish leukemic cells from normal lymphocytes and monocytes.The ratio of false postive was low and the results was reliable.
