烟叶生长和晾制期间细菌种群动态变化──烟草微生物细菌调控技术研究之一 被引量:12

摘要 为准确衡量烟叶不同生长期及晾制过程中不同时期细菌数量的变化,采用8个白肋烟品种对生产过程的细菌总数进行了连续分离计数。采集烟株封顶时的顶叶、收获成熟叶片及收获调制后3周和5周的烟叶样品,经表面灭菌后,用捣碎机捣碎。然后用0.1% Nacl系列稀释,涂布于TSA培养基上,在28~30℃的培养箱内分别培养24、48、72h后进行菌落观察和计数。结果表明,不同品种的细菌菌落在颜色、形态上的差异很大,细菌数量大约在干重102~106菌落形成单位(cfu)/g。在检测的2年中,1999年的细菌总数明显低于2000年,尤其在晾制过程中这一差别更为明显。 A better understanding of bacteria growth during tobacco curing may lead to improved tobacco quality during this process. Total bacteria were counted in 8 tobacco varieties during two consecutive growth seasons to quantify changes in bacterial population at different stages during growth and curing. Leaf tissue samples were collected at topping harvest 3 and 5 weeks after harvest in a conventional air-curing barn. Homogenized tissue was plated on growth media and the bacterial colonies developed were quantified after 2448 and 72 hours. Bacteria populations ranged from 102 to 106 cfu/g tissue sample. These populations were significantly lower in 1999 than 2000. In 2000 there were also significantly more bacteria during curing. There appeared to be no consistently significant differences in bacteria populations among the different tobacco varieties. This information may be useful to further investigate the curing process or different curing processes.
