摘要
用 40个随机引物 (1 0 -mers)对 2 2份魔芋资源材料的染色体组DNA进行PCR扩增 ,有 9个引物能在全部材料中扩增出 84条重视性好且稳定的DNA谱带 ,其中 77条具多态性。采用UPGA软件对扩增产物进行聚类分析表明 ,风岚野魔芋 (Amorphophallusspp)、兴仁黄魔芋 (A .spp)及罗悃魔芋 (A .spp)亲缘关系最近 ,花魔芋 (A .KonjacK .Koch)与白魔芋 (A .albusLiuetChen)关系次之 ,而东京魔芋 (A .tokinensisEngletGehrm)及 (A .dunniiTutcher)与花魔芋、白魔芋关系较远。根据聚类结果可把 2 2份魔芋材料划分为 5个组 。
The genomic DNAs isolated from 22 Amorphophallus species/accessions were used to analyze the genetic relationship among them by RAPD technique. Of 40 decamer nucleotide primers screened, 9 showed consistent banding patterns and amplifications in all the materials tested and produced 84 bands, of which 77 appeared to be polymorphic. The 22 species/accession were clustered into 5 groups by UPGA (unweighted pair-group average). The classifications tree showed that the topology of the tree portrayed the relationship among the species. Three successions cultivated in Guizhou were most closely related. A. konjac and A. albus were shown to be closely related genetically. A. konjac appeared to be the most distant species from A. tokinensis , followed by A. dunnii . Such results are believed to provide molecular biological information for sorting out Amorphophallus resources in China.
