富氧气体对低压缺氧大耳白兔肾EPO基因表达的影响 被引量:1

摘要 目的 :观察吸入不同浓度富氧气体对 110 0 0m低压条件下大耳白兔肾EPO基因表达的影响 ,从分子生物学水平为机载分子筛制氧系统的应用提供理论依据。方法 :2 4只大耳白兔随机分为 4组 (n =6 ) :空气对照组 ,6 3%富氧气体组 ,83%富氧气体组 ,纯氧组。将各组动物置于低压舱内 ,供给不同浓度的富氧气体 ,低压舱上升到 110 0 0m ,停留 30min ,然后将动物即刻处死 ,提取肾组织总RNA ,用RT -PCR方法检测肾组织EPOmRNA表达情况。结果 :大耳白兔暴露于 110 0 0m后 ,空气对照组EPOmRNA表达的阳性比率为 6 / 6 ,6 3%富氧气体组为 4 / 6 ,83%富氧气体组为 3/ 6 ,纯氧组为 1/ 6。随着吸入气氧浓度的增加 ,EPOmRNA表达的阳性率降低。吸氧浓度与EPO基因表达的阳性率呈明显负相关 (r =- 0 .976 0 4 ,P <0 .0 5 )。结论 :吸入较高浓度富氧气体对 110 0 Objective:To observe the effects of inhaling different concentrations of enriched oxygen on rabbit kidney EPO gene expression at 11 000 m hypobaria, so as from molecular biological level to provide theoretical basis for application of airborne molecular sieve of oxygen supplying system. Methods: 24 rabbits were divided randomly into 4 groups:air control group,63% enriched oxygen group, 83% enriched oxygen group and pure oxygen group(n=6).The animals were exposed to 11 000 m hypobaric hypoxia, in the meantime they inhaled different concentrations of enriched oxygen gas for 30 min. Then they were decapitated after the treatment. Total RNA was isolated by single-step acid guanidinium thiocyanate-phenol-chloroform extraction method. The mRNA expression of EPO in rabbit kidney were measured by RT-PCR. Result: The EPO mRNA expression ratio of air control group, 63% enriched oxygen gas group, 83% enriched oxygen group and pure oxygen group were 6/6, 4/6, 3/6 and 1/6 respectively . The EPO mRNA expression ratio in rabbit kidney decreased with the increasing of inhaled oxygen concentration. The inhaled oxygen concentration and the EPO mRNA expression ratio showed obvious negative correlation (r=-0.97604,P<0.05).Conclusion: Inhaling higher concentration of enriched oxygen could inhibit EPO mRNA expression in rabbit kidney at 11 000 m.
