摘要
目的 :探讨分离、培养大鼠主动脉平滑肌细胞 (VSMC)的方法 ,为研究损伤性血管疾病提供有用的体外研究模型。方法 :用组织贴壁、反转干涸的方法分离、培养大鼠主动脉平滑肌细胞并进行传代培养。采用形态学和抗α-SM -actin免疫组织化学鉴定证实为SVMC。结果 :细胞呈VSMC特征性“峰”“谷”状生长 ,历次原代培养经鉴定均为高纯度的VSMC。经传代培养至 2 0代 ,细胞维持原有生长性状 ,细胞活力未见减低。结论 :组织贴壁法培养大鼠主动脉平滑肌细胞具有简便、易行、细胞损伤小、活力及纯度高的优点 ,是研究损伤性血管疾病良好的体外研究模型。
Objective:To determine the technique of culture Rat aortas smooth muscle cell.Methods:Rat aortic smooth muscle cells(VSMCs) were prepared from aortas by modified explant technique and cells were passaged.The purity and identity of the VSMCs cultures were verified by morphology and using a monoclonal antibody against α-SM-actin immunohistochemistry studies.Results:High purity VSMCs were obtained.At confluence,these cells formed a hill-and-valley pattern typical of cultured VSMCs.The cell viability didn't decline after cell passaged to 20.Conclusions:Tissure explant method cultivation of rat VSMCs was small harmful to the cell viability and possessing high purity.This culture system provide a simple and convenient method for studying disease related to vascular injury.
