犬小肠缺血再灌注后氧自由基的改变意义 被引量:8

摘要 目的研究小肠小范围缺血再灌注后的氧自由基改变及其意义方法阻断分布于较小范围的小肠动脉,于动脉阻断前、阻断开放后0,30及60min,从与阻断动脉伴行小肠静脉采集血液标本.检测其 NO 和 SOD 的浓度.结果再灌注后,在0.30及60min 的 NO 浓度分别是(12.1±4.6),(11.6±4.4).(15.5±4.8)μmol·L^(-1),SOD 的浓度分别是(75.0±4.5),(43.4±1 1.4),(90.3±27.9)×10~3NU·L^1.NO 和 SOD 的改变特点显示小肠再灌注后其浓度明显低于阻断前(25.6±4.0)μmol·L^(-1)和(169.0±10.8×10~3NU·L^(-1),P<0.01).在再灌注30min 为最底,而至再灌注60min 则有明显的升高.结论小范围的小肠缺血再灌注而导致氧自由基升高和小肠损伤.与多脏器和大范围小肠缺血再灌注相比,虽然也很明显.但恢复较快. AIM To study the changes and implication of oxygen free radical of ischemia-reperfusion in limited area of the intestine. METHODS The intestinal ischemia-reperfusion model was established by clamping the artery distributing in small area,the blood samples were collected from accompaning intestinal vein before claming the artery and at 0,30 and 60min after reperfusion,the concentration of nitric oxide (NO) and superoxide dismutase (SOD) were examined. RESULTS After reperfusion,the concentration of NO at 0,30 and 60min was (12.1±4.6),(11.6±4.4),(15.5± 4.8) μmol·L^(-1),and that of SOD at 0,30 and 60min was (75.0±4.5),(43.4±11.4),(90.3±27.9)×10~3NU·L^(-1) respectively.The concentrations of NO and SOD decreased obviously compared with those before artery clamping (25.6+4.0) μmol.L and (169.0±10.8)×10~3NU ·L,(P<0.01),being the lowest at 30min reperfusion, but it increased at 60min of reperfusion. CONCLUSION Limited area ischemia-reperfusion of intestine could increase oxygen free radical and the damage of the intestine.Although the changes of oxygen free radical were obvious,and it restored quickly as compared with ischemia-reperfusion of more viscere or in larger area.
