摘要
目的 比较基因传感器表面两种探针固定法的优劣。方法 分别用巯基固定法及生物素 亲和素固定法将人类乳头瘤病毒 (HumanPapillomaVirus,HPV)的探针固定在基因传感器的金膜表面 ,比较不同探针浓度、不同靶序列浓度下反应所引起的频率的变化以及所用的时间。结果 两种固定方法检测靶序列的灵敏度相当。生物素 亲和素法反应所需的时间较短 ,且具有放大效应 ,能结合上更多的探针并检测到更多的靶序列 ,但操作较为繁琐。巯基法操作较为简单 ,但所需的反应时间较长。结论 两种探针固定法各有利弊 ,实际操作中应根据不同的需要选择合适的固定方法。
Objective To make comparison between the 2 methods for immobilizing probe on the surfaces of piezoelectric gene sensors. Methods Human papilloma virus (HPV) probe was immobilized on the gold surface of gene sensor array with thiol method and biotin avidin method, respectively. The changes of frequency and time cost were compared in reactions with different concentrations of probe and target sequences. Results The sensitivity of 2 methods was comparative. The biotin avidin method took less time to react with target sequence and immobilized more probe, and hence combined more target sequence, but with tedious procedures. While thiol method was simple but much more time costing for reaction. Conclusion The 2 immobilization methods both have their own advantages and disadvantages. Suitable method should be chosen in the practice according to different requests.
