摘要 目的 探讨胆囊收缩素 (Cholecystokinin ,CCK)促进胆管癌细胞增殖的可能机制。方法 采用细胞增殖计数法及流式细胞仪分析CCK对胆管癌细胞生长、增殖及细胞周期的影响 ,RT PCR、Western印迹法检测CCK对P2 1WAF1 、P2 7KIP1 基因表达的影响。结果 CCK具有显著促进无血清培养的胆管癌细胞的生长和增殖 ,加速细胞周期的进程 ,P2 1WAF1 、P2 7KIP1 mRNA和蛋白水平表达明显降低。结论 CCK促进胆管癌细胞增殖可能与下调细胞周期相关蛋白P2 1WAF1 、P2 7KIP1 Objective To study the effect of cholecystokinin (CCK) on the growth of cholangiocarcinoma cell line QBC939, and explore the relationship between P21 WAF1 and P27 KIP1 gene expressions and QBC939 cell proliferation. Methods Cell counting and flow cytometry (FLC) were used to investigate cell growth, proliferation and cell cycle of the QBC939 cells after co cultured with CCK. The expressions of P21 WAF1 and P27 KIP1 were determined with immunohistochemical staining. Results The proliferation of QBC939 cells with CCK treatment showed an increase in the percentage of cells at S phase and a concomitant reduction of cells at G 1phase, which could be inhibited by L60. The expression levels of P21 WAF1 and P27 KIP1 genes were markedly down regulated in response to CCK treatment. Conclusion The promotion of CCK to QBC939 cell proliferation may be related to the down regulation of P21 WAF1 and P27 KIP1 genes.
