摘要
目的 探讨D 半乳糖致衰老小鼠学习记忆与海马突触界面结构可塑性变化的关系。方法 用开场行为和Y 迷宫分辨学习记忆行为分别检测D 半乳糖衰老模型小鼠自发活动和学习记忆行为的变化 ,然后用电镜和计算机图象分析仪观测小鼠海马CA3 区GrayⅠ型突触界面结构参数的变化。结果 D 半乳糖致衰老小鼠在新异环境中的自发活动和探究行为明显减少 ,学习记忆功能显著降低 ;记忆保持率 (% )由 76 8± 9 3下降为 6 8 6±9 4(P <0 0 5 ) ;同时衰老小鼠出现海马突触后致密物质 (PSD)厚度明显变薄 (33 30± 6 5 8,P <0 0 1) ,突触间隙宽度显著增大 (2 0 32± 1 83,P <0 0 5 )等突触界面超微结构的改变。结论 D 半乳糖致衰老小鼠脑功能的减退可能与其改变海马突触界面超微结构有关。
Objective To study the Yelationship between the learning\|memory behavior and the hippocampus synaptic structure alteration of aging\|mice induced by D\|galactose.Methods The spontaneous behavior and the learning memory behavior were tested in aging mice using open field and Y\|maze.The structure of Gray I synaptic interface in the CA\-3 area of hippocampus was quantitatively analyzed by electron microscopy and computer image processing appliance.Results Compared with the control group,D\|galactose was showed to decrease significantly the spontaneous behavior and explorative response in the open field,and injure remarkablely the learning and memory ability of the mice.The percentage of memory retention decreased from 76\^8±9\^3 to 68\^6±9\^4( P <0 05).Meanwhile the thickness of post\|synaptic density(PSD) was reduced(33\^30±6\^58 nm, P <0\^01),the width of synaptic celft in the hippocampus CA\-3 area was widened(20\^32±1\^83 nm), P <0\^05).Conclusion D\|galactose showed an effect of memory impairment in mice.An alteration of synaptic interface structure in hippocampus of mice may be involved in the effect.
