用 3种方法 ,提取来自漳州的香蕉束顶病病叶组织中的 DNA,并以此为模板 ,用台湾大学及美国夏威夷大学的 4种引物进行 PCR检测 ,通过 2种反应成分、 3种循环参数的选择试验建立了 PCR反应体系 ,并发现用CTAB法提取到的模板 DNA无论质量、数量都较高 ,用于 PCR扩增 ,反应灵敏且效果好 ,可以从 5μg的香蕉病叶组织中检测到
Total DNA was extracted from BBTV\|infected banana leaves using three different methods, CTAB method was shown to be better than others, and the DNA templates with high quality and quantity could be obtained. A protocol suitable for identifying BBTV strains isolated from Fujian province was established through optimizing the reaction conditions, including different primers, reaction systems, PCR cycles and their combinations. Using the modified procedure, BBTV within 5μg infected tissue could be detected successfully.
