摘要
目的 探讨成人成骨细胞骨保护蛋白 (OPG )的表达与年龄变化的相关性。方法 体外分离培养 12例成人成骨细胞。年龄 39~ 74岁 ,大于 60岁以上和小于 60岁以下者各 6例。采用逆转录 聚合酶链方法 (RT PCR)检测不同年龄段OPGmRNA的表达。结果 大于 60岁以上组OPGmRNA表达的相对含量为 0 .2 8± 0 .10 ,小于 60岁以下组为 0 .49± 0 .14 ,其差异有显著性(P <0 .0 5 ) 。结论 成骨细胞分泌的OPG随年龄的增长而逐渐减少 ,有可能是增龄性骨丢失的重要原因。
Objective To investigate the relationsh ip between the age and the osteoprotegerin by human osteoblast cultured in vitro.Methods 12 cases of adult osteoblast were isolated and cu ltured in vitro.The age ranges from 39 years old to 74 years old.Two groups are divided by their age.Expression of mRNA transcripts of osteoprotegerin were asse ssed by RT-PCR.Results The expression of mRNA transcripts of osteoprotege rin of the younger group(<60 years old) was higher than that of the older group (>60 years old)(P<0.05).Conclusion Osteoprotegerin expression was declined with a ge,and this may contribute to the age-related bone loss.
