桂皮酸诱导白血病肝癌肺癌细胞的分化和抗肺癌细胞侵袭作用 被引量:4

摘要 【目的】探讨桂皮酸对人早幼粒白血病细胞 (HL 6 0 )、人肝癌细胞 (BEL 740 2 )、人肺癌细胞 (PGCL3)分化和抗PGCL3细胞侵袭的影响。【方法】用组织学方法观察各种癌细胞分化的形态学表现 ;用硝基四氮唑蓝 (NBT)还原试验定量测定HL 6 0细胞的机能分化 ;用放射免疫测定法 (RIA)测定肝癌细胞AFP分泌量 ,用酶学方法测定该细胞的鸟氨酸氨基甲酰转移酶 (OCT)、酪氨酸 α 酮戊二酸转氨酶 (TAT)、碱性磷酸酶 (ALP)和γ 谷氨酰转肽酶 (γ GT)的比活力 ;用层粘连蛋白粘附测定法、纤粘蛋白趋化迁移测定法和重建基底膜侵袭测定法测定肺癌细胞的粘附、运动和侵袭能力。【结果】 1mmol/L和 2 5mmol/L桂皮酸处理使HL 6 0细胞形态向成熟方向分化 ,NBT反应显著增强 (P <0 0 1和P <0 0 5 )。 1mmol/L和 3mmol/L桂皮酸处理使BEL 740 2和PGCL3细胞出现分化改变 ,使BEL 740 2细胞AFP分泌量和γ GT比活力显著下降 (P <0 0 5和P <0 0 1) ,反映肝癌细胞分化的OCT、TAT和ALP 3种酶的比活力则显著升高 (P <0 0 1和P <0 0 5 ) ,使PGCL3细胞软琼脂集落形成率下降 ,细胞的粘附能力下降 (3mmol/LP <0 0 1) ,运动和侵袭能力均显著降低 (P <0 0 0 1)。【结论】桂皮酸具有诱导HL 6 0、BEL 740 To investigate the effect of cinnamic acid (CA) on the differentiation of human promyelocytic leukemia cells (HL 60), human hepatocarcinoma cells (BEL 7402), human lung cancer cells (PGCL3), and the invasive ability of the PGCL3 cells. The morphological differentiation of the cancer cells was observed by histological technique. Functional differentiation of HL 60 cells was examined with NBT reduction test. The AFP secretary amount of the hepatocarcinoma cells were determined with radioimmunoassay (RIA), and the specific activities of ornithine carbamoyl transferase (OCT), tyrosine α ketoglutarate transaminase (TAT), alkaline phosphatase (ALP) and γ glutamyl transpeptidase (γ GT) were assayed by enzymological methods. The adhesion ability, the migrated ability and invasive ability were determined with the laminin adhesion test, the chemotactic migration test and the invasion test of reconstituted basement membrane. 1 mmol/L and 2 5 mmol/L of CA induced maturation in morphology, enhanced the NBT reaction (P<0 01 and P<0 05) of the HL 60 cells. 1 mmol/L and 3 mmol/L of CA induced maturation in morphology of the BEL 7402 cells and PGCL3 cells, decreased the secretary amount of AFP and the specific activity of γ GT (P<0 05 and P<0 01), increased the specific activities of OCT, TAT and ALP (P<0 05 and P<0 01), that represented differentiation of hepatocarcinoma cells, of the BEL 7402 cells. Decrease of the colony forming rate in the semisolid agar (P<0 001), adhesion rate (3 mmol/L P<0 01), the migrated ability and invasive ability (P<0 001) of the PGCL3 cells were also observed after CA treatment. [Conclusion] Our studies identified that CA could induced differentiation of HL 60 cells, BEL 7402 cells and PGCL3 cells, and could inhibit the invasive ability of the PGCL3 cells.
