摘要
目的 探讨皮肤创伤修复中uPA/uPAR的表达规律及意义。方法 应用免疫组化、WesternBlot、细胞培养、ELISA等方法检测小鼠皮肤伤缘和培养的单层角质形成细胞创伤模型的uPA/uPAR的表达。结果 创伤后uPA/uPAR的表达增高 ,伤后 1 2～ 2 4h为uPA/uPAR的表达高峰。结论 创伤促进了uPA/uPAR的表达 。
Objective To explore the rule of urokinase type plasminogen activator (uPA) and its receptor (uPAR) expressions and their significance in skin wound repair. Methods A total of 48 mice were inflicted to skin wound model by resecting a piece of skin of 0.5 cm in size on the back. Would cultured keratinocytes were obtain by scratching epidermal cells at passage 2 after isolation from 20 day fetal mouse back with metal probe. Immunocytochemistry(ICC), western blot and ELISA were used to observe the expressions of the activator and its receptor in the skin tissues 1 mm around the skin wound and in the would cultured keratinocytes. Results The expressions of uPA and uPAR were increased both in vivo and in vitro , and reached the peaks in 12 to 24 h after the resection or scratching. Conclusion uPA and uPAR participate in the regulation of the keratinocyte differentiation. The increase of uPA and uPAR exerts a beneficial effect on the skin wound healing and re epithelialization.
