摘要
目的 观察比较阿霉素 ( ADM)化疗与 43℃加热化疗对耐药人肝癌细胞模型 - 772 1 /Adm(以下简称 772 1 /Adm细胞 )的敏感性及细胞内药物浓度的影响。方法 以人肝癌细胞模型 -772 1 /Adm为研究对象 ,采用水浴加温法、体外细胞毒试验 ( MTT法 )、流式细胞技术 ,观察阿霉素( ADM)化疗与加热化疗后细胞的存活率及细胞内阿霉素浓度的变化。结果 ( 1 )阿霉素化疗、加热化疗 30、60 min后 772 1 /Adm细胞存活率分别为 70 .2 %、40 .8%和 60 .2 %、37.4% ;( 2 )流式细胞仪检测显示阿霉素化疗、加热化疗 30 min后细胞内阿霉素浓度分别为 41 .3%、92 .0 %。结论 加热可以显著对抗 772 1 /Adm的耐药性 ,提高其对阿霉素的敏感性 ,这与加热提高了细胞内药物浓度有关。
Objective To comparatively observe the effects of thermo\|chemotherapy of ADM on human multidrug\|resistant hepatocellular carcinoma cell line (7721/Adm) to sensitivity and intracellular concentration of ADM.Methods By means of water bath and MTT assay,the sensitivity of 7721/Adm to ADM was observed,and the effects of hyperthermia on intracellular concentration of ADM of 7721/Adm was detected by flow cytometry.Results(1) Cell line survive rates of 7721/Adm were 70.2%?40.8% after chemotherapy and thermo\|chemotherapy of ADM on in 30min,and that of 7721/Adm were 60.2%?37.4% in 60min.(2) intracellular concentration of ADM of 7721/Adm were 41.3%?92.0% after chemotherapy and thermo\|chemotherapy of ADM on in 30min,detected by flow cytometry.Conclusion Heat treatment can apparently confronte multidrug\|resistant of 7721/Adm and increase the sensitivity of 7721/Adm to ADM.,and the resulet indicate that increasing the sensitivity can be related to intracellular concentration of ADM.
