摘要
目的 探讨卡介苗细胞壁骨架 (BCG CWS)联合白细胞介素 2 (IL 2 )膀胱灌注对膀胱肿瘤细胞凋亡及端粒酶活性的影响。 方法 采用末端脱氧核苷酸转移酶介导缺口末端标记法(TUNEL)和端粒重复序列扩增法 (TRAP)分别检测 33例膀胱癌患者膀胱灌注前后肿瘤细胞凋亡及端粒酶活性。 结果 膀胱灌注前后肿瘤细胞凋亡指数分别为 (35 .6± 12 .7) %和 (79.1± 11.3) % ,灌注后肿瘤细胞凋亡指数明显增加 ,P <0 .0 1,差别有显著性意义。灌注前肿瘤细胞端粒酶活性强度“ -”3例、“ +”6例、“ ++”15例、“ +++”9例 ,灌注后“ -”10例、“ +”16例、“ ++” 6例、“ +++”1例 ,灌注后肿瘤细胞端粒酶活性明显降低 ,P <0 .0 1,差别有显著性意义。 结论 BCG CWS联合IL 2膀胱灌注能抑制肿瘤细胞端粒酶活性 ,诱导肿瘤细胞发生凋亡 。
Objective To study the cellular apoptosis and telomerase activity in bladder cancer before and after treatment with cell wall skeleton of bacillus Calmett Guerin(BCG CWS) and interleukin 2(IL 2). Methods The cellular apoptosis and telomerase activity of 33 cases of bladder cancer tissues were detected by terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling(TUNEL) and telomeric repeat amplification protocol(TRAP) assay both before and after treated with BCG CWS and IL 2. Results The apoptosis index for bladder cancers before and after treatment with BCG CWS and IL 2 was (35.6±12.7)% and (79.1±11.3)% respectively, the apoptosis index being obviously increased after the treatments, P <0.01.The telomerase activity before treatment was “-”in 3,“+” in 6,“++” in 15 and “+++” in 9 whereas it was “-” in 10,“+” in 16,“++” in 6 and “+++” in 1 after the treatment.The intensity of telomerase activity decreased obviously after the treatment ( P <0.01). Conclusions BCG CWS and IL 2 is effective in inhibiting the telomerase activity and in inducing cellular apoptosis for bladder cancer.
