摘要
目的研究离体培养时瘤菌根菌属(Epulorhizasp.)真菌AR-18对铁皮石斛和金钗石斛生长的影响。方法采用琼脂作支持物,考察真菌对铁皮石斛和金钗石斛的鲜重和干重的影响;采用蛭石作支持物,考察真菌和营养液对铁皮石斛和金钗石斛的鲜重和干重的影响。结果在琼脂培养基上,真菌AR-18对铁皮石斛和金钗石斛的鲜重和干重的增加均没有显著性影响(P>0.05)。在蛭石基质上,真菌AR-18对金钗石斛鲜重的增加具有显著促进作用(P<0.05),接菌的金钗石斛的鲜重较不接菌提高16%;真菌AR-18对金钗石斛干重的增加具有极显著促进作用(P<0.01),接菌的金钗石斛的干重较不接菌提高21%。在蛭石基质上,真菌AR-18配合使用蒸馏水极显著促进了金钗石斛鲜重的增加(P<0.01),接菌的金钗石斛的鲜重较不接菌提高47%,真菌AR-18配合使用营养液对金钗石斛鲜重的增加没有显著性影响(P>0.05);真菌配合使用营养液或配合使用蒸馏水对金钗石斛干物质的积累没有显著性影响(P>0.05),所以最佳处理组合为真菌AR-18配合使用蒸馏水。在蛭石基质上,真菌AR-18和营养液对铁皮石斛的鲜重和干重的增加没有显著性影响(P>0.05)。结论在离体培养条件下选择合适的基质和控制真菌的适度生长是建立稳定的共生平衡系统和促进金钗石斛生长的重要条件。在人工栽培金钗石斛的?
Objective To examine effects of fungus AR-18(Epulorhiza sp.)on the growth of Dendrobium can didum and D.nobile in vitro culture.Methods Effects of fungus AR-18on fresh weight and dry weight of D.candidum and D.nobile were studied in vitro culture,when agar was used as rest,and effects of fungus AR-18and nutrients on fresh weight and dry weight of D.candidum and D.nobile were studied in vitro culture,when vermiculite was used as rest.Results In agar med ium,eff ects of fungus AR-18on fresh weight and dry weight of D.candidum and D.nobile were not sig-nificant (P>0.05).However,in vermiculite medium,the effect of fungus AR-18on fresh weight of D.nobile was significant (P<0.05).Fresh weight of D.nobile inoculated with fungus AR-18was in creased by16%compared with the non-inoculated ones(control).The effect of fungus AR-18on dry weight of D.nobile was very significant (P<0.01).Dry weight of D.nobile inoculated with fungus AR-18was increased by21%compared with the control.In vermiculite medium,the effect of combination treatment (Fungus AR-18+Distilled H 2 O)on fresh weight of D.nobile was very significant (P<0.01).Fresh weight of D.nobile inoculated with fungus AR-18was increased by47%compared with the control,while the effect of combination treatment (Fungus AR-18+Nutrients)on fresh weight of D.nobile was not significant (P>0.05).In addition,the effect of combinations treatment (Fungus AR-18+Nutrients)and(Fungus AR-18+Distilled H 2 O)on dry weight of D.nobile was not significant (P>0.05).So the combination treatment (Fungus AR-18+Distilled H 2 O)was the best for the growth of D.nobile.In vermiculite medium,effects of fungus AR-18and nutrients on fresh weight and dry weight of D.candidum were not significant (P>0.05).Conclusion It is important in vitro culture to select a proper rest for the control of the growth of fungus to establish a beneficial symbiotic relation-ship between plants and fungi.If the fungus AR-18is utilized as biological manure in got-up planting of D.nobile,a good result may be obtained.
