摘要
采用强酸型阳离子树脂为交换剂的离子交换柱 ,和经组氨酸基团修饰的强碱型阴离子树脂为交换剂的离子交换层析柱串联 ,以氯化钠溶液为洗脱剂 ,对透明质酸粗品进行分离和纯化。得到的透明质酸精品的蛋白质含量低于 0 .0 75 % ,平均分子量大于 9.41× 10 5,纯化收率为 5 8%~ 6 1%。并对洗脱剂的流速、浓度、p H等条件进行比较 ,确定了最佳洗脱条件。
Successful purification of hyaluronic acid (HA) was achieved in about 60% recovery by a strong acid cation exchange resin column which was connected i n series with another strong base anion exchange resin (modified with histidin e ) column, using NaCl solution as an eluant. The molecular weight of the purifie d HA was more than 9.41×10 5 and the protein content was less than 0.075%. The i nfluences of flow rate, concentration, and pH value of eluant upon purification were studied to give the optimum condition.
