摘要
目的 探讨热休克对卵巢癌细胞A2 780经TNF—α诱导凋亡的影响 ,为卵巢癌细胞凋亡与热休克相关关系提供实验依据。方法 采用脱氧核苷酸末端转移酶介导的DUTP缺口末端标记法 (TUNEL)检测细胞凋亡指数 (AI) ,3H—TdR掺入试验检测DNA合成情况。结果 热休克组凋亡指数比非热休克组的凋亡指数明显下降 ,差异有显著性意义 ,且呈剂量依赖效应 ,cpm值热休克组细胞比非热休克组明显增高 ,抑制率下降 ,也呈剂量依赖效应。结论 热休克对卵巢癌细胞A2 780凋亡具有抑制作用 。
Objective To study the effect of HS for ovarian cancer cell line A2780 apoptosis induced by TNF-α with a different density,and provide basic material in correlationship between apoptosis and HS of ovarian cancer cells.Methods TUNEL was used to exam apoptosis index(AI).At the same time, 3H-TdR test was used for examing DNA synthesis.Results The apoptosis index of treated HS group has decreased more significantly than those untreated by HS group,with significant effect depending on the dose of THF-α.Simultaneously,the cpm of treated HS group has more significantly increased than the one of untreated HS with depending on the dose of TNF-α.Conclusion HS can inhibit the apoptosis induced by TNF-α in ovarian cancer cell in vitro,which depends on the dose of TNF-α.
