摘要
目的 :研究菝葜的活血化瘀药理作用。方法 :菝葜水煎液 ( )、正丁醇萃取物 ( )、正丁醇萃取剩余物 ( )高 ( 1 0 0 g/kg)、低 ( 5 0 g/kg)剂量对小白鼠灌胃给药 7d后 ,眼眶取血 ,采用凝固比浊法测定 APTT、PT,采用 derive法测定 FIB;全自动血细胞计数仪测定血小板数 ,采用比浊法体外测定 、 、 对人血小板的聚集功能 Ma、M1 、V1 的影响。结果 :1 0 0 g/kg剂量的 和 能显著延长 APTT( P<0 .0 5 ) ,5 0~ 1 0 0 g/kg剂量的 能使 FIB显著增加 ( P<0 .0 5 ) ,而 5 0 g/kg剂量的 能使 FIB显著减少 ( P<0 .0 5 )。稀释 8倍后的 和 能抑制体外的血小板聚集功能 ,特别是对 V1 的影响更显著 ( P<0 .0 1 ) ,但三者对血小板数无明显影响。结论 :菝葜具有明显活血化瘀的药理作用 ,其作用机制可能与抑制血小板聚集和延长内源性凝血时间及影响纤维蛋白原生成有关。
AIM The purpose is to study the pharmacological action of the extracts from Smilax china L. on promoting blood circulation to remove blood stasis. METHODS ig the high (100 g/kg) and low(50 g/kg) dose of the extract with water from Smilax china L.(I),the extract with n-butanol from Ⅰ (Ⅱ)and the remainder(Ⅲ) to mice and drawing blood from eyepit after seven days. Activated- partial thromboplastin time (APTT), prothrombin time(PT) were measured by turbimetry method and fibrinogen(FIB) was determined by derive method. Platelet aggregation M 1,Ma,V 1 was measured by turbimetry method in vitro. RESULTS The high dose of I,Ⅲ can prolong APTT obviously(P<0.05), Ⅲ can increase FIB obviously and Ⅱ can decrease FIB(P<0.05). They all have no influence on PT and the number of platelet.Ⅰ and Ⅱ (0.84 g/ml)can inhibit the platelet aggregation, especially influence on the velocity of aggregation (V 1) (P<0.05) . CONCLUSIONS %Smilax china% L. has the pharmacological action of promoting circulation to remove blood stasis. The mechanism may be connected with inhibiting the platelet aggretion, prolonging the clotting time and influencing on the synthesis of FIB.
