摘要
目的寻找一种能够适合临床需要的较为理想的皮肤替代物。方法采用组织工程技术,将种子细胞接种于复合壳多糖真皮基质,先后进行沉没培养和器官型培养,观察培养物变化。结果获得的人工皮肤有较好组织学特性和物理学特性。结论复合壳多糖人工皮肤组织学特点接近天然皮肤,有可能成为一种新型的皮肤替代物,应用于临床。
Objective To investigate an ideal skin equivalent for the need of clinic.Methods According to the methods of tissue en-gineering,fibroblasts taken from h uman foreskin were casted in the derm al matrix gel mixed with chitosan,a n atural cationic polysaccharide,typeⅠcollagen of rat tail' s tendon,concentrated Dulbecco,s M odified eagle,s medium(DMEM)and glycosaminoglycan(GAG),which then was seeded with keratinocytes to pro duce a biolayered skin equivalent under aseptic condition.After submering culture in keratinocyte culture medium for 1week,the artificial ski n was raised to the air-liquid interface by stainless steel mesh for anoth er week in laboratory.Results The composite chitosan artificial skin we achived had excellent handling ch aracteristics with elasticity,flexibility and tenacity.Histolog ic exami-nation revealed the composites comp rised of epidermis and dermis.Conclusion It is suggested that the composite ch itosan artificial skin may be a novel skin equivalent applied in clinic in the future for its histolog ic feature similar to that of nature s kin.
